I am wondering this as well. Since the DLCs make up ~ 70% of the game's size, does one benefit from learning the base game first and then expanding?

Can the DLCs be turned on and off at launch, or once installed, is it irremovably a part of the game? Thanks!

I saw in the release thread someone say you can choose what parts of the dlc you want on or off.
If you guys are brand new to the game then I suggest you play through the in game tutorials to get acquainted with everything.

With regards to customization, the game is very granular. You can turn on/off entire expansions, types of ships, factions, and ai behaviors. If you want to just jump into a game then you should just play against the "vanilla" ai type with everything under AI Plots and AI Options tabs set to off. There are some things there that are just an advantage with little to no downsides, but having everything off would make for the least complicated game.

Don't worry too much about the difficulty. There are like 15 or something difficulties that gradually ramp up the pain. You can set it up so that the ai is morally opposed to harming you, or so that you lose in 5 minutes. For a first time game, it's really up to you. Difficulty 7 is "normal", but I find it pretty challenging. Maybe you would want to start on 5 or less?
Definitely play the tutorials first. They're long. Do them anyway. This is not Starcraft, it's a genre blend that's evolved over the last 5 years, and it takes some getting used to.

Next baby step: play a "Beginner" script game. Start scripts are an option in the game lobby. Win your beginner game or lose it. Leave the default options on for this, or fiddle with a few of them.

Next game, just turn a few DLC features on. You can leave the DLC's all enabled, just experiment with a few of the lobby options. There are a lot of them, so don't get overwhelmed. Just pick 3-6 that you're interested in. Or play the base game if you need to practice fundamentals.

Alternatively, turn them all on and experience the onslaught!

They're not SquareSoft long, but they are around four/five hours in total.

It's also important to note these tutorials are not the hit-button-get-bacon kind of tutorial where you have no choice but do whatever the tutorial says. You want to ignore the computer's advice (note: advice, not orders) and play your way, you can. Of course since this is your first time play you will almost certainly lose hard, but that's also learning.
